Efficient evolution of human antibodies from general protein language models
Plasmids were transformed into Stellar competent cells , and transformed cells were plated and grown at 37 °C overnight. Colonies were mini-prepped per the manufacturer’s recommendations and sequence confirmed and then maxi-prepped per the manufacturer’s recommendations . Plasmids were sterile filtered using a 0.22-μm syringe filter and stored at 4 °C.All proteins were expressed in Expi293F cells .
CoV-2 Spike, the samples were then incubated at 4 °C while stirring overnight. CoV-2 Spike antigens were incubated at room temperature while stirring overnight. Resin/supernatant mixtures were added to chromatography columns for gravity flow purification. The resin in the column was washed with wash buffer , and the proteins were eluted with 250 mM imidazole, 20 mM HEPES pH 7.4, 150 mM NaCl or 20 mM imidazole, 1× PBS.
